A DGK isoform, which resides in the nucleus of PC12 cells, is DGK-theta. Here, we show that nerve growth factor (NGF) treatment of serum-starved PC12 cells results in the stimulation of both a cytoplasmic and a nuclear DGK activity. However, time course analysis shows that cytoplasmic DGK activity peaked earlier than its nuclear counterpart.

Juli 2017.

(1998) concluded that the nuclear localization of DGK-iota is regulated by phosphorylation in much the same manner as that of DGK … 2020-10-10 Previous evidence has shown that the nucleus contains several DGK isoforms. In this study, we have analyzed the expression and subnuclear localization of DGK‐ζ employing C2C12 mouse myoblasts. Immunocytochemistry coupled to confocal laser scanning microscopy showed that both endogenous and green fluorescent protein‐tagged overexpressed DGK‐ζ localized mostly to the nucleus. Nuclear DGK-zeta increased during myogenic differentiation of C2C12 cells, while DGK-zeta down-regulation by siRNA markedly impaired differentiation.

2007-01-01 · The list of subcellular compartments where DGK activity or DGKs have been found includes, but is not limited to, the plasma membrane, the endoplasmic reticulum, the neuromuscular junction, the cytoskeleton, and even the nucleus. In fact, within the nucleus, DGKs appear to be confined to separate, distinct regions (Topham and Prescott, 2002). Indeed, the NES mutant forms of DGK-ζ accumulated in the nucleus to a much greater extent than wild type DGK-ζ. Moreover, treatment with leptomycin B, an inhibitor of leucine-rich type NES, resulted in accumulation of both endogenous and ectopically expressed DGK-ζ in the nucleus, demonstrating that nuclear export of DGK-ζ is chro- We found 15–30% of total cellular DGK-ζ in the nucleus both of COS-7 cells transfected with DGK-ζ complementary DNA and of A172 cells, a glioblastoma-derived line, which express native DGK-ζ .

Further fractionation of the nuclei can be achieved using protocols that have been described ( Ledeen and Wu, 2004 ). ABSTRACT The nucleus contains diacylglycerol ki-nases (DGKs), i.e., the enzymes that, by converting diacylglycerol (DG) into phosphatidic acid, terminate DG-dependent events.

One DGK, DGK-zeta, contains both a nuclear localization signal and nuclear export signal , suggesting a shuttling between the nucleus and the cytoplasm. Morphological studies on animal tissues report that DGK-zeta predominantly localizes to the nucleus in some cell types, while it exhibits both nuclear and cytoplasmic localization in others.
